Note that early plant responses may be dose-dependent. Products with moderate concentration of AM fungi are not expected to show positive growth effects after short times of 2-3 weeks, as the fungi need some time to establish symbiosis, propagate, and promote plant growth. Very early symbiotic stages may even show transient growth depression, reflecting the carbon-invest of the plant.

As international customer have very variable demands regarding the administrative processes, we are not setting up a web-based system for scientific customers. Products

We have reduced our portfolio to the Rhizophagus irregularis isolate that is used as a model organism in the vast majority of research approaches.

Our research-grade AM fungal inoculum contains mainly fungal spores as propagules (a very low number of small root fragments may be present). All material is produced aseptically, in vitro. We supply AMF propagules in diatom earth ('diatomite') powder with 50% humidity content. If you need material for larger scale research attempts (e.g. field experiments), please inquire for more economical but less well defined standard-grade inoculum (root fragments and spores, not packed under aseptical conditions). Specific formulations like in liquid or in gel may be produced and delivered on demand, but this may take several weeks or months.

Carrier material 50 g
The material is identical to the carrier in the powder based products, but contains no water (thus, apply half the weight when compared to the inoculum, for control experiments). It can, e.g., be use for control experiments or for dilution of inoculum. Only sold to SYMPLANTA inoculum customers. Material is gamma-irradiation sterilized, but re-packed and sealed under non-sterile conditions.

Species monitoring - NGS (MiSeq) based

Please inquire well in advance, as we may need up to 3 months for individual projects, dependent on sample numbers and sequencing availablity.
We may request a 50% pre-payment.

DNA based high throughput species identification 'MiSeq' from various (field soil, roots, culture substrate, etc.) samples.
PCR in triplicate, pooling, 450-500 bp based identification of AM fungi at species level, using a well established marker in the LSU rRNA gene region.

Species identification - DNA based

DNA based species identification 'basic' from individual spores or roots (material provides must be from an isolate [single species]).
Identification of AM fungi based on PCR product (amplicon) sequencing, one sequence of 450-500 bp of the D2 region of the LSU rRNA gene, PCR amplicon consensus sequence including nucleotide polymorphisms. This fragment usually is sufficient for species identification using our database (provided the species is characterized). A maximum likelyhood phylogenetic analysis and a phylogenetic tree of the respective genus including the query sequence is included.

Inoculum viability and efficiency tests

Bioassay for quantification of the % root length colonization (RLC, in %) efficiency of substrates, products (e.g., commercial inocula), or soil.
Based on a standard procedure (inoculum application in 3 or 4 dilutions, 5 replicates each, root staining, % root length colonization analyses 5-7 weeks after inoculation) using a selected lettuce variety as test plant. If you need testing of other target plants, please inquire. Note that we will need about 8 weeks for the entire procedure.

Inoculum viability and efficiency tests

Standardised test of the % of viable spores in substrates, products (e.g., commercial).
Based on a standard procedure (MTT staining, 4 replicates, estimating number of putatively viable spore content). Note: this is only applicable with suited substrates (microscopic counting has to be possible) and also suited AMF species. Also note, that MTT positive reaction is not in all cases a 100% proof vor viability - if you need to be sure about colonization potential, you should apply a bioassay. MTT can be efficiently used for process control and QC of defined formulations.

Species identification - DNA based

DNA based species identification 'detailed', from individual spores or mixed material (spores, roots, soil, ...).
PCR, cloning, RFLP screening, and DNA sequencing based identification of AM fungi, including 10 diverse >1500 bp sequences.
Inquire for complex samples, which e.g. demand multiple initial PCR reactions (pooled before cloning), and screening of larger clone numbers (approx. additional costs 100 €, but depends on number of sequences to be provided).

AM fungal spore abundance is determined by spore extraction from 50 ml soil, taxonomic identification, and counting of the different morphotypes.
